It works by increasing fluid secretion in the intestines and accelerating bowel transit, providing relief from constipation and abdominal discomfort.

Mechanism of Action

• Linaclotide binds to guanylate cyclase-C (GC-C) receptors on intestinal epithelial cells.

• This increases cyclic GMP levels, which:

  • Activates CFTR ion channels, leading to secretion of chloride and bicarbonate into the intestinal lumen.

  • Increases intestinal fluid and electrolyte secretion, softening stools.

• It also reduces visceral pain by modulating intestinal sensory nerve activity.
