Leuconamo contains Leucovorin Calcium, a reduced form of folic acid (vitamin B9). It is used as a chemoprotective agent to reduce the toxic effects of methotrexate and to enhance the effectiveness of 5-fluorouracil (5-FU) in cancer therapy. It is available in oral tablets or injectable forms and is also used to treat folate deficiency.
Mechanism of Action

Leucovorin Calcium is converted into tetrahydrofolate, which participates in DNA synthesis and repair.

With methotrexate: Leucovorin bypasses methotrexate-induced dihydrofolate reductase inhibition, rescuing normal cells from toxicity.

With 5-FU: It enhances binding of 5-FU to thymidylate synthase, increasing its anticancer efficacy.
Uses

Rescue therapy after high-dose methotrexate.

Enhances 5-fluorouracil (5-FU) chemotherapy in colorectal and other cancers.
